The antiproliferative effect of MNBE on cervical cancer cell lines. (A) HeLa, SiHa, ME-180, and non-cancerous L2 cells (104 cells/well) were seeded into a 96-well plate, incubated overnight, and then treated with the 0–300 µg/ml of MNBE for 24 h. Viability was measured by MTT assay, and the viability of untreated cells was set at 100%. Results graphed using the log (inhibitor) vs. response—Variable slope (four parameters) curve using GraphPad Prism. (B) HeLa, SiHa, and ME-180 cells were seeded in 60 mm dishes in a range of 200–1000 cells and allowed to grow for 24 h. Cells were then treated with corresponding IC25 and IC50 doses of MNBE for 24 h. After treatment, media was replaced with a fresh medium, and cells were incubated for 10–14 days. Cells were fixed in 4% paraformaldehyde for 20 min and stained with 0.4% crystal violet for 15 min. The stained cells were viewed and counted under a microscope. All experiments were performed in triplicate, and data are expressed as the mean ± SEM of the three independent experiments.
