Tan IIA rescued cell viability and inhibited apoptosis via activating MEK/ERK1/2/GSK-3β pathway. (A) Cell viability was assessed by MTT assay; (B) reactive oxygen species (ROS) production; (C) superoxide dismutase (SOD) activity; (D) Annexin V-FITC/PI staining, as measured by flow cytometry, revealed a significant increase in the apoptotic cell ratio in the H2O2 treatment group, and this increased apoptosis was reduced by Tan IIA. (E) Tan IIA increased the expression of p-MEK, p-ERK1/2 and p-GSK-3β, as detected by immunoblotting. G15 could diminish the expression of these proteins induced by Tan IIA.
