Cucurbitacins inhibit nerve growth factor (NGF)-mediated tropomyosin receptor kinase A (TrkA) pathway in PC12 cells. The inhibition of TrkA phosphorylation induced by cucurbitacin B (CuB), CuE, and CuI was assayed in the presence or absence of the indicated cucurbitacin concentrations, and the inhibitor GW441756 as a positive control. (a) CuB inhibited TrkA and downstream Akt phosphorylation in a concentration-dependent manner. (b) CuE inhibited TrkA and downstream Akt phosphorylation in a concentration-dependent manner. (c) CuI inhibited TrkA and downstream Akt phosphorylation in a concentration-dependent manner. The blot shown is representative of at least three experiments
