Regulation of SIRT1 and FOXO1 gene by SAL in HUVECs exposed to ox-LDL. HUVECs were pretreated with SAL (20, 50, 100 μM) for 2 h, and then incubated with ox-LDL (100 μg/mL) for 48 h. qRT-PCR shows that the mRNA levels of SIRT1 (a) and FOXO1 (b) were significantly reduced by ox-LDL, but was increased by SAL in a concentration dependent manner. Western blotting was performed to determine protein levels of SIRT1 (c) and FOXO1 (d). In HUVECs with SAL (100 μM), cells were pre-incubated with FOXO1 inhibitor (AS1842856, 100 nM), or SIRT1 inhibitor nicotinamide (NAM, 200 nM). SAL significantly attenuates the decrease in SIRT1 (e) and FOXO1 (f) proteins in HUVECs exposed to ox-LDL.
