LERD reversibly inhibits antigen-stimulated degranulation in mast cells. RBL-2H3 mast cells (A) and BMMCs (B) were incubated overnight in 24-well cluster plates with 50 ng/ml of IgE in a complete growth medium. The IgE-primed mast cells were stimulated by antigen (25 ng/ml) with or without LERD. The activity of β-hexosaminidase was measured as described in the “Methods” section. (C) RBL-2H3 cells were pre-incubated for 1 h with 0.1% DMSO or 100 μg/ml LERD. After LERD-treated cells were washed five times with PIPES buffer, degranulation of mast cells was measured as for panel A. (D) BMMCs were incubated with LERD for 8 h and the cell viability, then, was measured as described in the “Methods” section. The mean ± SEM from three independent experiments are shown here. Asterisks indicate significant differences from the controls, *P < 0.05 and **P < 0.01. PP2 is a general Src family kinase inhibitor.
