Inhibition of TNF-α-induced MMP-9 mRNA and protein expression by SSeo in HASMCs. (A) Total RNA was isolated from cells grown under the same conditions as Figure 1 and reverse-transcribed. Resulting cDNAs were then subjected to PCR. The reaction products were run on 1% agarose gel electrophoresis and visualized by EtBr staining. (B) The cells were sampled, lysed, and 30–50 μg of protein was separated by SDS-polyacrylamide gel electrophoresis. Western blotting was then performed using the indicated antibodies and an ECL detection system. GAPDH and actin were used as the internal controls for the RT-PCR and Western blot analyses, respectively.
