PPC enhanced the IFNγ + /CD8 + T cell response to a DNA vaccine specific for ovalbumin. PPC enhanced the DNA vaccine induced production of OVA-specific IFNγ+/CD8+ T cells. Mice were immunized by DNA injection followed immediately by electroporation. Mice were injected with DNA encoding full-length ovalbumin and then the injection area was immediately exposed to 95 volts in four pulses of 65 milliseconds using an Electro Square Porator device (BTX, model TX830). CD8+ T cells were isolated from the splenocytes population and plated with stimulator cells (EL4 only, EL4 pulsed with the OVA257 peptide, or EG7 cells). Mice receiving no PPC are represented by the open bars while those receiving 200 μg/mL PPC ad libitum are represented by the filled bars. Error bars represent the standard deviation. The delivery of PPC was found to significantly enhance the number OVA specific IFNγ+/CD8+ T cells (p = 0.009 for EL4 OVA257 and p = 0.005 for EG7).
