Figure 4From: Polygonum viviparum L. induces vasorelaxation in the rat thoracic aorta via activation of nitric oxide synthase in endothelial cells Effect of Polygonum viviparum (PV) on endothelial nitric oxide (NO) synthase (eNOS) phosphorylation, cytosolic nuclear factor E2-related factor 2 (Nrf2) degradation, and heme oxygenase (HO)-1 activation. (A) Human umbilical vein endothelial cells (HUVECs) were treated with PV (at the indicated concentrations) for 0.5 or 24 h. Total proteins (30 μg) were analyzed on an 8% SDS-PAGE with an antibody specific of eNOS Ser-1177; and analyzed on 10% SDS-PAGE with an antibody specific of Nrf2 and HO-1. (B) The relative density was calculated as the ratio of eNOS (phosphor form) expression to eNOS (total form) expression. In addition, the relative density was calculated as the ratio of Nrf2 degradation and HO-1 expression to β-actin expression. Each electrophoretogram is representative of the results from more than 5 individual experiments. * p < 0.05, # p <0.05, ξξ p < 0.01, significantly different from the control.Back to article page
